In a nutshell, formalin fixation occurs by the reaction of formalin with the primary amines; the subsequent cross-linking of tissue proteins with formalin gives rise to formation of methylene bridges. Methylene bridges fix and preserve tissue antigens. Retrieval techniques are attempts to reverse formalin fixation by breaking methylene bridges and expose as many antigenic sites for primary antibodies to bind and, therefore, enhance the staining signals.

High Heat Retrieval (HIER)

HIER techniques are arbitrary and non-standardized practices with many variables.1-3 Deparaffinized tissue slides are immersed in varied buffers with different pHs and heated at boiling or slightly sub-boiling temperatures for 10-30 minutes. Slides then undergo a long cooling period before primary antibodies are applied. The selections for HIER heating time, buffers, pHs and heating devices are selected arbitrarily for each given primary antibody by both antibody vendors and IHC labs. It is important to note that the very high heat is the major factor in breaking the methylene bridges in HIER technique and the buffers used play a nominal role.

HIER, pH and Temperature Interplay

pH and temperature are inversely proportional. As the temperature rises, the pH drops. This phenomenon is the basis for the use of many HIER solutions with varied pHs. The two most commonly employed are citrate buffers pH 6 (acidic buffer) and high pH buffers pH 8-10 (alkaline buffer). The use of home-brewed solutions are also common and a minor variation on the basic theme.

Citrate buffer pH 6 was the original HIER buffer; high pH buffer arrived later. Citrate buffer pH 6 drops to pH 2-3 above temperature of 90o and higher, and such acidic pH is detrimental to the survival of many tissue antigens. The use of high pH buffer starting at pH 8-10 is an attempt to compensate for the drastic drop in pH associated with high heat in the HIER technique.

The high pH buffer is mainly used to provide a more neutral pH environment for tissue sections during the intense and pressurized heating period associated with HIER. Most tissue degradation and morphology damage in HIER are generated by extreme heat alone. EDTA added to high pH buffer has no effect on retrieval and no scientific merit. EDTA is a chelating agent; it may work well for bone tissues chelating the calcium but has no enhancing effect on retrieval or staining. 		<description>Charles Taze Russell did not believe in such an organization as "Jehovah's Witnesses", the leaders of which have often misrepresented Russell. Russell was a non-sectarian, who did not believe that the church needed a authoritative organization such as the organization that Joseph Rutherford developed after Russell's death.  Rutherford then rejected the very core doctrine that Russell had defended, that is, the ransom for all.

Russell gave a summation of his beliefs, what he stood for, in the January 15, 1912 issue of the Watch Tower, page 28:

I. We affirm the humanity of Jesus and the deity of Christ.

II. We acknowledge that the personality of the Holy Spirit is the Father and the Son; that the Holy Spirit proceeds from both, and in turn from all who are begotten by it.

III. We affirm the resurrection of Christ--that He was put to death in the flesh, but quickened in the Spirit. We deny that He was raised in the flesh, and challenge any statement to that effect as being unscriptural.

IV. We affirm, with the Scriptures, that God alone possessed immortality, "dwelling in the light which no man can approach unto." We affirm that this Divine quality has already been granted to the Lord Jesus and is to be the portion of the elect Bride, the "Body of Christ." As for mankind, we affirm the Divine provision for these [R4956 : page 29] and for angels to be everlasting life for the obedient. This, by many, is mistermed immortality. We follow the Scriptures strictly.

V. We hold that the entire race lost life with Father Adam, as a result of his failure in Eden; and that Christ died to secure a second chance for Adam and an individual chance for all of his race, who lost their first chance in Adam when he sinned. "As all in Adam die, even so all in Christ shall be made alive." (I Cor. 15:21,22.) A few of us, comparatively, having eyes of faith and ears of understanding, have had this second chance in the present life. Adam and the great mass of his posterity must get their second chance after being awakened from the tomb. But NOBODY IS TO GET A THIRD CHANCE!

VI. We believe that the soul was condemned to death. "The soul that sinneth it shall die." We believe that this death would have been eternal, everlasting destruction, had it not been for God's mercy in and through our Lord's redemptive work. By reason of His death our souls do not die in this full sense of the word, but are Scripturally said to "fall asleep," "asleep in Jesus." The awakening will be in the resurrection morning; and the interim will be a period of unconsciousness, beautifully symbolized by a restful sleep.

VII. We believe in the "hell" of the Bible, sheol. This, the only word used for hell for four thousand years, is translated more than one-half the time grave in our Common Version, and should always be thus translated. "Hades," in the New Testament, is its equivalent. "Gehenna fire," of the New Testament, is a symbolical picture declared to signify the Second Death.

VIII. We believe that God is able to destroy "both soul and body" in Gehenna--the Second Death. We consider it much more sane to believe thus, as it is more Scriptural, than to believe that in creating man God did a work which He could not undo; much more reasonable also than to believe He prefers to have the incorrigible suffer eternally, when their sufferings could do neither themselves nor others any good.

IX. We believe that, like the Father and the holy angels, our Lord is a spirit being. We are convinced that "flesh and blood cannot inherit the Kingdom of God." We do not believe that our Lord has a flesh and blood body, a "little lower than the angels," and has thus been out of harmony with His heavenly environment for nearly two thousand years. We believe the Apostle's statement, "Now the Lord is that Spirit." This is the Jesus who will "so come, in like manner," quietly and unknown to the world, as He went away. We do not affirm, dogmatically, that He came in 1874, but we say that to us it is the evident teaching of the Scriptures. Our Lord warned us not to expect Him in the flesh; that men might say "Lo, here" or "Lo, there." The Harvest work in the universal Church Nominal, the Laodicean period of the Church, well corroborates our expectations of what His work will be, as outlined in His parables, etc.</description>

The word 'Spiritualist' is therefore definitely of or relating to or connected with Spiritualism.

A spiritualist is not someone who necessarily is able to act as an intermediary between the spirit world and the physical world; that is an ability that a Spiritualist medium has. They cannot act as an intermediary between the living and the dead because those in spirit are not dead. They are intermediaries between those living in the spirit dimensions and those living in the physical dimension.

It is not correct to state that a spiritualist is is an intermediary nor is it correct to describe 'mediums' as Spiritualists unless it is clearly specified that they are Spiritualist mediums. There are many people who call themselves mediums who are not connected in any way with Spiritualism. The correct use of the word medium is: a Spiritualist who is able to communicate with the spirit world. Many who currently use the world are not able to do this nor do they attempt to do so. They are mere psychics with no necessary spiritual or religious affiliation or interest.</description>

		<description>I really think that the principles and concepts that are used in macrobiotic practice could be useful to a broader base of people.
The idea that people are going to wake up one day and decide to have miso soup and steamed greens is not exactly realistic.
It seems that people find macrobiotics either because they have two weeks to live or they are into other areas of natural health / healing for various reasons , but the majority of people are not privy to this philosophy.</description>
